[TYPO3] Can't execute PHP-files in typo3conf or any of its subfolders!

Hi
I can't execute PHP-files in the typo3conf folder or any of its 
subfolders. And because of this I can't use any local extension :-(

All the files and libs permission settings are set to 775.
What should I do?
