Let me counter your question with one of my own,

"When I get into my car it will not start even though I turn the key - 
I've looked at the servicemanual but I must have missed something"

Not really that easy to tell what's wrong with this little information, 
no? ;-)

What does happen when you attempt to put an item in the basket, what pid 
are you refering to and is the commerce mod correctly configured on that 
page? etc, etc
