I asked this question a month or two ago, but didn't get any hard answers.
As a result I tried both ways -- hosted several sites under one
installation, and also individual installations.
The "one-installation" approach has the benefit that you only need to import
and configure plugins once. The main drawback is that sync'ing sites between
a development server and a production server is pretty-well impossible once
the client starts changing and modifying their own site (if there's a way
around this, please let me know!) The biggest benefit is that you can "whip
up" a site in a few minutes. I have several sets of templates prepared, and
I can sit down with a client and build the site in front of their eyes --
makes it a little harder for them to say "no". ;-)
Individual sites require more involvement in setting things up, which can
add several hours to the development process. I'll be getting around this by
creating my own distribution ("Cthulhu3"), which I'll begin selling online
as soon as I remove all of those pesky copyrights*. Our hosting provider
provides a skeleton directory where we can pre-install any software. When we
create an account, the CMS is ready and waiting (almost -- still need to
create the DB). Almost all of the sites we do involve multiple languages,
and a bunch of other features, so we'll be packaging everything together so
we can start-off more quickly. I think this is the best trade-off.
I would be interested in hearing others' solutions to running multiple
sites. We do around two sites a month, and are expecting a big increase in
the near future.

Our templates have an XHTML validation link at the bottom. Sometimes it
passes, sometimes it doesn't -- depends what extensions are being used.
*This is a joke. Chill.
