I guess you are just starting to setup a new site and haven't worked much
with TYPO3 before or?
There are several ways to start learning more about TYPO3.
One would be starting from scratch and do intensive developing and trying to
understand every step .... takes a loooonnnng time!

and download and install the template you like - you can also download
several templates and than compare how it is done. Learning by example on a
RUNNING system.

How to modify a template is described here

If you only want to modify your template so that it looks like i.e. your
HTML or Photoshop Mokup, than use this

It is very fast to achieve your goal, very flexible and best of all it is
well documented! and very understandable!
