If this error comes in backend, it sounds to me it is not template 
based. The backend (all but Web/View-module) works either you have the 
typoscript template or not. The template is needed when you want to show 
the content in front end.

Are you sure you have proper rights to the BOTH folders and files? You 
have to open (drwxr-xr-x) to apache user the folders and files into 
which TYPO3 writes: fileadmin, typo3conf, typo3temp and uploads.
