Copy the entire site folder/file tree & mysql typo3 database to a backup 
folder/CD/tape/etc....use microsoft's backup utility to create a .bkf

actually, you don't need to back up the entire folder tree...you need 
/fileadmin, /typo3, /typo3conf, /typo3temp, /uploads
