There's a part of it that tells what you need. Yes, you'll need an
extension, so you can create your own preset:

$GLOBALS['TYPO3_CONF_VARS']['RTE']['Presets']['my_preset'] =
'EXT:my_layout_extension/Configuration/RTE/Default.yaml';

And then define the preset you want for every RTE you have:

RTE.config.tx_news_domain_model_news.bodytext.preset = default

You can also override the default preset (
$GLOBALS['TYPO3_CONF_VARS']['RTE']['Presets']['default']).

I don't know if you can do it without an extension, only using typoscript.
