I have so many pesonal extensions.
This means it will surely not benefit Typo3 in the short run for a lot 
of people want to upgrade,
however I do think it will benefit Typo3 in the long run.

Typo3 5.0 will take at least 2 years to develop and I would suggest 
start over from scratch setting up the new
fundamet for Typo3 core using PHP version 5. (I hope 6 will not be out 
by then :) ). All sorts of items can be re-thought over
from DBAL to Typoscript.

We must at least support Typo3 version 4.x for the 4 years to give 
people. While the guru's can start working on a Typo3 version 5.
Maby to even speedup development we need to start thinking of doing a 
fund-raising for mayor core devs.
With such a large userbase and big company's using it I am sure we can 
start doing some serious marketing and start a fundrasing?

I am all for a complete refactored Typo3 Version 5 and which we can at 
least import a 4.x branche.
